Characteristics human mesangial cell from a 16-weeks-old female fetus with 36 h after spontaneous abortion; primary cell obtained from Clonetics Corp. (San Diego, CA) and later immortalized with a SV40 large T antigen expression plasmid.
Biomaterial provider Transformed cell kindly provided by Dr. Bernard Banas, from Nephrological Center, Medical Policlinic, Ludwig-Maximilians University, Munich, Germany.
Treatment protocol cells were cultured in serum-containg media until confluence, when cells were kept for 24 hours in DMEM 0.5% FBS. Cells were stimulated with 10% FBS or serum free-medium for 6 hours.
Growth protocol Cells were grown in low-glucose Dulbecco’s modified Eagle’s medium (DMEM) supplemented with 10% fetal bovine serum (serum), containing 2 g/L sodium bicarbonate and antibiotics at pH 7.4

Extraction protocol QIAGEN RNeasy Mini Kit
Label streptavidin-Cy5
Label protocol Two micrograms of total RNA was labeled according to manufacturer's protocol (CodeLink Expression Analysis Tutorial Version 4.0).
 
Hybridization protocol Ten micrograms of fragmented target cRNA was used for hybridization of each UniSet Human I Expression Bioarray chip as described in manufacturer's protocol (CodeLink Expression Analysis Tutorial Version 4.0).
Scan protocol Processed slides were scanned using an Axon GenePix Scanner with the laser set to 635 nm, 600V, 100% power laser and the scan resolution to 10 µm.
Description RNA extracted from 3 culture dishes was pooled, producing one sample. A total of three samples (9 culture dishes) were hybridized for each group (1 samples/array).
Data processing Slides were scanned using CodeLinkTM Expression Scanning Software and images for each slide analyzed using the CodeLinkTM Expression Analysis Software and Microsoft Excel.
