RB1-depleted osteoblast response to ionizing radiation: time course

Analysis of RB1-depleted primary osteoblasts exposed to 4 Gy of ionizing radiation (IR) for up to 24 hours. IR exposure and germline mutations in RB1 are risk factors for developing osteosarcoma. Results provide insight into the role of RB1 in the response of osteoblasts to IR.

Post-slippage cells increase expression of factors associated with SASP.

SKP2 knockout in Rb1/p53 deficient transgenic mouse models of osteosarcoma induces macrophage infiltration and drives transcription of genes with favorable prognosis in patients

(Submitter supplied) To study SKP2 in osteosarcoma, we generated a Skp2 KO line and a Skp2 intact line of transgenic OS mouse models: germline Skp2 knockout (“TKO”: Osx1-Cre;Rb1lox/lox;p53lox/lox;SKP2) versus Skp2 intact (“DKO”: Osx1-Cre;Rb1lox/lox;p53lox/lox). We then performed bulk RNAseq of the SKP2 KO vs SKP2 intact OS.

Screening bone marrow derived murine mesenchymal stem cells for tumourigenicity

(Submitter supplied) To study early events in osteosarcoma genesis murine bone marrow derived mesenchymal stem cells (mMSCs) were studied which generated osteosarcoma-like tumours upon injection into nude mice. Loss of the retinoblastoma tumor suppressor is a common event in Basal-like and Luminal B breast carcinomas

(Submitter supplied) Breast cancers can be classified using whole genome expression into distinct subtypes that show differences in patient prognosis. One of these groups, the basal-like carcinomas, are poorly differentiated, highly metastatic, and genomically unstable. These tumors also contain specific genetic alterations with one example being frequent p53 mutations. Identification of conserved gene expression features across human and murine mammary tumors

(Submitter supplied) Mice have been used as models for human breast cancers for many years, however, it is still unclear which murine models faithfully represent human tumor phenotypes. To address this question, we used DNA microarrays to characterize 10 different murine mammary models and compared these data to the expression patterns from primary human breast tumors.
