Gene expression profiling of intracranial aneurysms

(Submitter supplied) The rupture of intracranial aneurysm (IA) causes subarachnoid hemorrhage associated with high morbidity and mortality. We compared gene expression profiles in aneurysmal domes between unruptured IAs (UIAs) and ruptured IAs (RIAs) to elucidate biological mechanisms predisposing to the rupture of IA. We determined gene expression levels of eight RIAs, five UIAs, and 10 superficial temporal arteries (STAs) with the Agilent microarrays. more...

Comparison of gene expression between ruptured and unruptured human intracranial aneurysms

(Submitter supplied) Background and Purpose Aneurysmal subarachnoid hemorrhage, almost always from saccular intracranial aneurysm (sIA), is a devastating form of stroke that affects working age population. Cellular and molecular mechanisms predisposing to the rupture of the sIA wall are largely unknown. Such knowledge would facilitate the design of novel diagnostic tools and therapies for the sIA disease. Methods We compared the whole genome expression profile of eleven ruptured sIA wall samples, resected at a median of 15 hours after rupture, to that of eight unruptured ones. more...

RNA Sequencing Analysis of Intracranial Aneurysm Walls Reveals Involvement of Lysosomes and Immunoglobulins in Rupture

(Submitter supplied) Background and Purpose—Analyzing genes involved in development and rupture of intracranial aneurysms can enhance knowledge about the pathogenesis of aneurysms, and identify new treatment strategies. We compared gene expression between ruptured and unruptured aneurysms and control intracranial arteries. Methods—We determined expression levels with RNA sequencing. Applying a multivariate negative binomial model, we identified genes that were differentially expressed between 44 aneurysms and 16 control arteries, and between 22 ruptured and 21 unruptured aneurysms. more...

Identification of microRNA as novel circulating markers for presence of intracranial aneurysms

(Submitter supplied) We carried out a case control study in an attempt to identify changes in circulating microRNAs in patients with intracranial aneurysms (IAs). We selected 40 cases (20 ruptured and 20 unruptured) and 20 healthy controls. We randomly selected 5 samples from each group and combined them into a sample pool. In this way we obtained 12 sample pools and one pool was used for a single microarray. Changes in microRNA levels in the plasma were surveyed with Agilent Human microRNA Microarray (Release 14.0, 8x15K). more...

Whole blood gene expression profiles of patients with a past aneurysmal subarachnoid hemorrhage

(Submitter supplied) Investigation of gene expression profiles in blood of previous aSAH patients, aiming to gain insight into the pathogenesis of IA and aSAH, and to make a first step towards improvement of aSAH risk prediction. The results indicate that no gene expression differences are present in blood of previous aSAH patients compared to controls, besides one differentially co-expressed gene network without a clear relevant biological function.

Expression data from intracranial arteries and intracranial aneurysms

(Submitter supplied) Gene expression information is useful in prioritizing candidate genes in linkage intervals. The data can also identify pathways involved in the pathophysiology of disease. We used microarrays to identify which genes are expressed in either intracranial arteries (control) or in intracranial aneurysms (case), and can therefore contribute to the disease phenotypes. We used microarrays to identify the pathway membership of expressed genes and the overrepresentation of pathways with expressed genes in the known linkage intervals for intracranial aneurysms. more...

Intracranial aneurysm associated single-nucleotide polymorphisms alter regulatory DNA in the human circle of Willis

(Submitter supplied) Background and Purpose: Genome-wide association studies significantly link intracranial aneurysm (IA) to single-nucleotide polymorphisms (SNPs) in six genomic loci. To gain insight into the relevance of these IA associated SNPs, we aimed to identify regulatory regions and analyze overall gene expression in the human circle of Willis (CoW), on which these aneurysms develop. Methods: We performed chromatin immunoprecipitation and sequencing for histone modifications H3K4me1 and H3K27ac to identify regulatory regions, including distal enhancers and active promoters, in post mortem specimens of the human CoW. more...

Lack of cyclic stretch disturbs endothelial cell transcriptomic profile

(Submitter supplied) Cyclic circumferential stretch (CCS) induced by pulsatile blood pressure affects endothelial cell (EC) gene expression and regulates vascular mechanical properties. Although alterations in EC gene expression in response to increased CCS as observed in hypertensive patients is widely studied, the EC response to low or lack of CCS as observed in intracranial aneurysm (IA) disease has been poorly investigated. more...
