Zhang et al. (2000) cloned THYN1, which they designated HSPC144, from CD34 (142230)-positive hematopoietic stem/progenitor cells. The deduced protein contains 225 amino acids.
Miyaji et al. (2002) cloned mouse Thyn1, which they called Thy28. The deduced 226-amino acid mouse protein shares 90% and 92% identity with human and chicken THY28, respectively. Human and mouse THY28 contain N-terminal nuclear localization signals and a putative tyrosine phosphorylation site. THY28 orthologs were also detected in bacteria, yeast, and plant. Sequence analysis revealed 4 regions of conservation among THY28 orthologs, with complete conservation of the tyrosine within the putative tyrosine kinase phosphorylation site of mouse and human THY28. Northern blot analysis detected variable expression of a 1-kb transcript in all mouse tissues examined.
Zhang et al. (2000) determined that the THYN1 gene contains at least 8 exons.
By radiation hybrid analysis, Zhang et al. (2000) mapped the THYN1 gene to chromosome 11q25.
Miyaji et al. (2002) mapped the mouse Thyn1 gene to a region of proximal chromosome 9 that shares homology of synteny with human chromosome 11q25.
