Clock mutant cecum response to Salmonella enterica serovar Typhimurium infection at different circadian times

Analysis of cecum from master circadian regulator Clock mutants infected with S. Typhimurium at two circadian times (10am and 10pm). Clock mutants infected at night exhibited higher inflammation. Results provide insight into the role of the circadian clock in the host immune response to infection.

Expression data from mouse cecum

(Submitter supplied) To adapt the lives of organisms to the day-night cycle, evolution has built a complex machinery, whose molecular components are able to anticipate and drive changes in organism behavior and metabolism. A mutual bidirectional interaction exists between circadian abnormalities and development of diseases. Microarrays were used to determine whether circadian regulation occurs at the transcriptional level in response to Salmonella Typhimurium infection.

Mouse Salmonella Study

(Submitter supplied) Salmonella enterica serotype Typhimurium causes an acute inflammatory reaction in the cecum of streptomycin pre-treated mice. We determined global changes in gene expression elicited by serotype Typhimurium in the cecal mucosa. The gene expression profile was dominated by T cell derived cytokines and genes whose expression is known to be induced by these cytokines. Markedly increased mRNA levels of interferon (IFN-gamma), interleukin-22 (IL-22) and IL-17 were detected by quantitative real-time PCR. more...

PARP1 and Salmonella

(Submitter supplied) BACKGROUND & AIMS: The immune system comprises an innate and an adaptive immune response to combat pathogenic agents. The human enteropathogen Salmonella enterica serovar Typhimurium invades the intestinal mucosa and triggers an early innate pro-inflammatory host gene response, which results in diarrheal disease. Several host factors are involved in the acute early response to Salmonella infection. more...

Expression data from non-infected and Salmonella Choleraesuis infected mesenteric lymph nodes

(Submitter supplied) To understand the host transcriptional response to S. enterica serovar Choleraesuis (S. Choleraesuis), the first generation Affymetrix porcine GeneChip® was used to identify differentially expressed genes in the mesenteric lymph nodes responding to infection at acute (8 hours (h), 24h, 48h post-inoculation (pi)) and chronic stages (21 days (d) pi) The objectives of this study were to identify and examine the stereotypical gene expression response within the host mesenteric lymph nodes to S. more...

Peripheral blood transcriptome responses to Salmonella Typhimurium in pigs differing in Salmonella shedding phenotypes

(Submitter supplied) Salmonella species infect many vertebrate species, and pigs colonized with Salmonella enterica serovar Typhimurium (ST) are usually asymptomatic, making detection of these Salmonella-carrier pigs difficult. The variable fecal shedding of this gram-negative bacteria in such pigs is an important cause of foodborne illness and zoonotic disease. To investigate gene pathways and biomarkers associated with the variance in Salmonella shedding following experimental inoculation, we have initiated the first analysis of the whole blood transcriptional response induced by Salmonella. more...

Early and Late gene expression responses to Salmonella infection in the mouse intestine

(Submitter supplied) We used microarrays to detail the global programme of gene expression of the mouse colon responses to Salmonella (SL1344 and SB1117) infection at the early phase (8 hours) and the late phase (4 days). We specifically examined the the differentillay gene expression profiles in mouse colon when it responded to pathogenic Salmonella stain SL1344 (with AvrA expression) or SB1117 (without AvrA expression).

Transcriptional responses of mouse BMM and TEPM to lipopolysaccharide (LPS)

(Submitter supplied) Evolutionary change in gene expression is generally considered to be a major driver of phenotypic differences between species. We investigated innate immune diversification by analyzing inter-species differences in the transcriptional responses of primary human and mouse macrophages to the TLR4 agonist, LPS. Using a custom platform permitting cross-species interrogation coupled with deep sequencing of mRNA 5’ ends, we identified extensive divergence in LPS-regulated orthologous gene expression between humans and mice (24% of orthologs, http://www.macgate.qfab.org). more...
