Dendritic cell responses to proinflammatory TNF cytokine or pathogenic Trypanosoma brucei antigens

Analysis of dendritic cells (DCs) treated with TNFa cytokine or T. brucei-derived, variant-specific surface glycoproteins (AnTat1.1 mfVSG or MiTat1.5 sVSG). Results provide insight into the partial maturation signatures of the differentially stimulated DCs.

Dendritic cell maturation by proinflammatory TNF or pathogenic Trypanosoma brucei antigens instruct similar T helper-2 cell responses in murine models of autoimmunity and asthma

(Submitter supplied) Background Dendritic cells (DC) represent the major cell type leading to polarized T helper (Th) cell responses in vivo. The current data indicate that DC maturation under inflammatory conditions or by helminth infections induce only incomplete DC maturation bearing a partially tolerogenic potential. Such semi-mature DC promoted a Th2 profile with increasing proportions of regulatory IL-10 producing T cells (Tr-1) upon restimulation. more...

Gene expression changes in C57Bl/6 and CD16-/- murine bone-marrow derived dendritic cells (BMDCs) following overnight stimulation with OVA or OVA-immune complex

(Submitter supplied) Atopic asthma is a chronic inflammatory disease of the lungs that is commonly associated with a Th2 response. The role of allergen-specific IgG in the initiation and development of allergic airway inflammation is still poorly understood; however, a receptor of IgG-immune complexes, CD16, has been demonstrated to promote augmentation of Th2 responses. To identify what genes downstream of CD16 signaling may be contributing to development of a Th2 response, we use ovalbumin (OVA) as our model antigen and compared wildtype and CD16-/- BMDCs that were treated overnight with OVA or OVA-immune complex.

Th2 responses are primed by skin dendritic cells with distinct transcriptional profiles

(Submitter supplied) Dendritic cell subsets purified from skin-draining lymph nodes of C57BL/6J mice following intradermal injection of non-viable Nippostrongylus brasiliensis (Nb) or topical application of Dibutyl Phthalate-FITC. Results provide insight into the genetic programme induced in DC exposed to Th2 stimuli.

Transcription factor C/EBPβ orchestrates DC maturation and functionality under homeostatic and malignant conditions [Emu-Myc]

(Submitter supplied) Dendritic cell (DC) maturation is a prerequisite for the induction of adaptive immune responses against pathogens and cancer. Transcription factor (TF) networks control differential aspects of early DC progenitor versus late stage DC cell fate decisions. Here, we identified the TF C/EBPβ as a key regulator for DC maturation and immunogenic functionality under homeostatic and lymphoma-transformed conditions. more...

Microarray analysis of therapeutic (GM4) and non therapeutic (GM) NOD DC

(Submitter supplied) We have previously demonstrated that bone marrow-derived DC can prevent diabetes development and halt progression of insulitis in NOD mice, the mouse model of type 1 diabetes (T1D). The DC population that was most effective in this therapy had a mature phenotype, expressed high levels of costimulatory molecules and secreted low levels of IL-12p70. The protective DC therapy induced regulatory Th2 cells that shifted the dominant Th1 environment, present in NOD mice, to a mixed Th1/Th2 milieu. more...

Single-cell transcriptomic analysis of mouse lung CD11b+ dendritic cells post allergic sensitization

(Submitter supplied) Conventional dendritic cells (cDCs) in the lung that drive effector T cell differentiation, and initiate allergic responses upon inhalation of allergens. However, since CD11b+ cDCs are heterogeneous, the specific function of each subpopulation has been elusive. To identify subpopulations of CD11b+ cDCs, we performed single cell RNA-sequencing (scRNAS-Seq) of total lung CD11b+ cDCs following inhalation of house dust extracts, and detected multiple clusters based on their differential transcriptomes. more...
