Lung metastatic breast cancer cell line response to metadherin depletion

Analysis of LM2 breast cancer cells depleted for metadherin (MTDH), a cell surface protein in breast tumors that mediates lung metastasis. LM2 cells were grown alone or on a monolayer of lung microvascular endothelial cells. Results provide insight into molecular mechanisms underlying MTDH action.

Cytoplasmic MTDH associates with mRNAs

(Submitter supplied) To examine whether MTDH is a novel RNA binding protein and regulates either metabolism or translation of various mRNAs, we performed a RNA-binding protein immunoprecipitation (RIP) with MTDH and IgG antibodies, and the resulting immunoprecipitated RNA was subjected to a microarray to identify transcripts associating with MTDH. In addition we tested the effect of PI3K inhibition using BEZ235 (a dual PI3K/mTOR inhibitor) on the association of MTDH with target mRNAs.

Hepatocyte growth factor effect on Met receptor-knockout primary hepatocytes: time course

Analysis of Met-deficient hepatocytes treated with HGF for up to 24 hours. HGF/Met signaling controls invasive growth and metastasis in a variety of cancers, including hepatocellular carcinoma. Results identify a Met-dependent gene expression signature that may help elucidate the oncogenic process.

Integrated genomic and function characterization of the 8q22 gain

(Submitter supplied) Integrated DNA and expression array analysis in primary human breast tumors identified chromosome 8q22 copy number gain and a suite of over-expressed genes in this region highly relevant to subsequent recurrence. 8q copy gain is associated with increased risk of distant metastasis despite adjuvant chemotherapy and multiple genes from 8q22 are overexpressed and have pleiotropic effects, contributing to tumor growth, survival, and reduced killing of tumor cell by chemotherapy.
