TAF7L is an X-linked germ cell-specific paralogue of TAF7, which is a generally expressed component of TFIID. Here we report the generation of Taf7l mutant mice by homologous recombination in embryonic stem cells using the Cre-loxP strategy. Taf7l cooperates with Trf2 to regulate spermiogenesis

(Submitter supplied) Taf7l (a paralogue of Taf7) and Trf2 (a TBP-related protein) are components of the core promoter complex required for gene/tissue-specific transcription of protein-coding genes by RNA polymerase II. Previous studies reported that Taf7l knockout mice exhibit structurally abnormal sperm, reduced sperm count, weakened motility and compromised fertility. Genetic characterization of a missense mutation in the X-linked TAF7L gene in an oligozoospermic man

(Submitter supplied) We carried out genome-wide expression profiling by RNA-seq to determine whether the mutation brings about a certain influence to the transcriptomic homeostasis. Indeed, there were some differential expressions of mRNA and noncoding genes in comparison of the Taf7lD144G and wide-type testis, lending support to mutation-caused defects at molecular level.

Dual Functions of TAF7L in Adipocyte Differentiation

(Submitter supplied) The diverse transcriptional mechanisms governing cellular differentiation and development of mammalian tissue remains poorly understood. Here we report that TAF7L, a paralogue of TFIID subunit TAF7, is enriched in adipocytes and mouse white fat tissue (WAT). Depletion of TAF7L reduced adipocyte-specific gene expression and compromised adipocyte differentiation as well as WAT development. Murine Histone Methyltransferase Mll5, is involved in Male Gametogenesis

(Submitter supplied) Male mice but not female Mll5 -/- mice are infertile. This study showed that post-meiotic spermatogenic maturation is impaired in Mll5 -/- mice. In order to investigate the role of Mll5 in spermatogenesis, a transcriptome analysis of whole testes from Mll5 knock out and wildtype mice was performed. Affymetrix Mouse Exon chips were hybridized with testes material from three individual male mice from each wild-type or Mll5 -/- genotype, in order to identify gene regulation differences attributed to the loss of Mll5.

Analysis of effect of sirt1 deficiency on global gene expression in testis

(Submitter supplied) Testis were obtained from 2 sibling pairs of of young male mice, 1 knock-out and 1 heterozygous for the Sirt1 allele, and used to compare global gene expression using spotted oligo microarrays (the MEEBO arrays from the Stanford microarray core- MOE series).
