Identification and validation of novel ERBB2 (HER2, NEU) targets including genes

(Submitter supplied) V-erb-b2 erythroblastic leukemia viral oncogene homolog 2 (ERBB2; synonyms HER2, NEU) encodes a transmembrane glycoprotein with tyrosine kinase-specific activity that acts as a major switch in different signal-transduction processes. ERBB2 amplification and overexpression have been found in a number of human cancers, including breast, ovary and kidney carcinoma. Our aim was to detect ERBB2- regulated target genes that contribute to its tumorigenic effect on a genomewide scale. more...

Activation of ER-alpha network in MMTV-Wnt/ILK mammary tumors

(Submitter supplied) A transgenic mouse model, MMTV-Wnt/ILK, with mammary specific expression of both Wnt-1 and ILK, was generated by crossing the two mouse lines MMTV-Wnt-1 and MMTV-ILK. Affymetrix Mouse Exon chips were hybridized with material from four independent mammary tumors from each MMTV-Wnt-1 and MMTV-Wnt/ILK transgenic model, in order to identify possible signaling differences involved during increased tumor incidence.

Distinct ErbB-2-coupled signalling pathways promote mammary tumors with unique pathological and transcriptional profiles

(Submitter supplied) ErbB-2 overexpression and amplification occurs in 15 - 30% of human invasive breast carcinomas associated with poor clinical prognosis. Previously, we have demonstrated that four ErbB-2/Neu tyrosine-autophosphorylation sites within the cytoplasmic tail of the receptor recruit distinct adaptor proteins and are sufficient to mediate transforming signals in vitro. Two of these sites representing the Grb2 (Neu-YB) and Shc (Neu-YD) binding sites can induce mammary tumourigenesis and metastasis. more...

ChIP-seq of Tcfap2c in Neu mammary cancer cells

(Submitter supplied) Tumors were harvested from mice carrying the rat activated Neu-oncogene and made into a single cell suspension. ChIP-SEQ was carried out as previously described in Kulak MV, Cyr AR, Woodfield GW, Bogachek M, Spanheimer PM, Li T et al. Transcriptional regulation of the GPX1 gene by TFAP2C and aberrant CpG methylation in human breast cancer. Oncogene 2013; 32: 4043-4051. Tcfap2c and RNA Pol II peaks can be found on their respective tracks.

Transcriptional Regulation of the GPX1 Gene by TFAP2C and Aberrant CpG Methylation in Human Breast Cancer

(Submitter supplied) The complexity of gene regulation has created obstacles to defining mechanisms that establish the patterns of gene expression characteristic of the different clinical phenotypes of breast cancer. Transcription factor TFAP2C plays a critical role in the regulation of both estrogen receptor-alpha (ERα) and c-ErbB2/HER2 (Her2). Herein, we performed chromatin immunoprecipitation and direct sequencing (ChIP-seq) for TFAP2C in four breast cancer cell lines representing different clinical phenotypes. more...

Effects of acute Notch activation on the mammary epithelial compartment in vivo

(Submitter supplied) Notch signaling is widely implicated in mouse mammary gland development and tumorigenesis. To investigate the effects of acute activation of Notch signaling in the mammary epithelial compartment, we generated bi-transgenic MMTV-rtTA; TetO-NICD1 (MTB/TICNX) mice that conditionally express a constitutively active NOTCH1 intracellular domain (NICD1) construct in the mammary epithelium upon doxycycline administration.

Identification of breast cancer tumor associated antigens

(Submitter supplied) Four prototypic situations, corresponding to progressive stages of mammary carcinogenesis, were analyzed. Total RNA from whole-mount for 10, 15, 19 and 22 weeks old BALB-neuT animals was extracted. Differential expression between the various prototypic conditions indicates the presence of changing in level of mRNAs due to the increment in the tumor mass (Astolfi et al. Cancer Immunol Immunother 2005, 54:599-610). more...

Mammary tumors from K14-cre; ApcCKO/+ mice vs control mammary glands

(Submitter supplied) Many components of Wnt/β-catenin signaling pathway also play critical roles in mammary tumor development. To study the role of Apc in mammary tumorigensis, we introduced conditional Apc mutations specifically into two different mammary epithelial populations using K14-Cre (progenitor) and WAP-cre (lactaing luminal) transgenic mice. Only the K14-cre mediated Apc heterozygosity developed mammary adenocarcinomas demonstrating histological and molecular heterogeneity, suggesting the progenitor cell origin of these tumors. more...
