miRBase is an RNAcentral consortium member that provides a searchable database of published miRNA sequence and annotation, as well as providing a nomenclature service to the miRNA community. Entries in this project include those assembled from public next generation datasets (ENA TPA TSA).
Available small RNA deep sequencing datasets were obtained from SRA and GEO. Small RNA sequencing reads were mapped to the miRBase 20 hairpin sequences using Bowtie (v 0.12.7), using a seed length of 17 (-l 18) allowing 1 mismatch (-n 1). After normalising each dataset by the number of reads mapping to all microRNAs, the most abundant read from each arm of each microRNA hairpin was chosen as the representative mature microRNA sequence.
